大肠杆菌-枯草芽孢杆菌穿梭质粒表达载体pHT43是Mo Bi Tec 公司的产品。在pHT01的基础上,将编码α-淀粉酶的amyQ基因的信号肽序列与pHT01的SD序列融合,构建成pHT43。pHT43质粒是高拷贝的穿梭质粒,全长8057bp,筛选标记为氨苄青霉素和氯霉素抗性。用IPTG诱导后,重组菌株能高水平分泌表达重组目的蛋白,并可分泌到培养基中。pHT43质粒的图谱和序列如下:

      @张阳阳 pHT01和pHT43质粒都可以在细胞质中高水平表达重组蛋白,其中pHT43载体可引导重组蛋白分泌到培养基中。
      pHT01的衍生载体带有不同的标签,其中pHT08带8×His标签,pHT09带链球菌标签(Strep标签),pHT10带C-Myc标签。
